Tissue Sample
Most fixatives should be satisfactory.
Protocol
- Bring sections to water via xylene and ethanol.
- Place into the staining solution for 15 minutes at 50°C.
- Rinse briefly with distilled water.
- Differentiate until staining is clear.
- Dehydrate with amyl alcohol.
- Clear with xylene and mount with a resinous medium.
Expected Results
- Nuclei – violet
- Plasma cell cytoplasm – red
- Other cell cytoplasm – pink
